Part:BBa_K1913005
lux quorum sensing system + GFP reporter
This part allows for lux system quorum sensing and visualizes quorum sensing with GFP expression.
Usage and Biology
This part contains the genes luxI and luxR originating from Vibrio fischeri. These genes allow gram-negative bacteria to communicate cell density through production and reception of 3-oxo-hexanoyl-homo-serine-lactones. These 3-oxo-hexanoyl HSLs induce GFP expression in this part to visualize the quorum sensing.
Figure 1. Fluorescence and absorbance data for E. coli quorum sensing strains. Continuous line: Fluorescence/OD600; Dashed line: Absorbance at OD600; Red: 2 plasmid quorum sensing strain; Green: 1 plasmid quorum sensing strain; Purple: reporter plasmid only strain. For both strains every value displayed is the average of at all technical replicates (at least three). All Fluorescence/OD600 data were normalized to the range of the 2 plasmid quorum sensing strain data which reached the highest value. The Fluorescence/OD600 data for the 1 plasmid quorum sensing strain were corrected to start at 0 (as they would otherwise have been negative). After almost 12 hours, the fluorescence from the 2 plasmid strains reached an overflow. Therefore no values after this point are displayed.
