Human Gut Hormone Glp1 (7-37) with upstream signal peptide PhoA
Same as part BBa-K1848001 except possessing upstream signal peptide PhoA. This signal peptide shuttles the target protein into the extracellular space via the type II secretion pathway. This signal peptide is E. coli specific and might not work well within other species of gram negative bacteria.
Building off of NTU-Taida 2012 iGEM's interest in the PhoA signal peptide and Glp-1, Columbia NYC iGEM aimed to express levels of secreted protein without needing extreme levels of concentration by working within a known probiotic E. coli strain (Nissle) more readily known to express high levels of protein secretion and expression without untoward infringment upon cellular metabolism. To this end we were successful for our blot represents a mere 10x concentrate supernatant compared to their 100x.
Below is a blot characterizing this part. The left column of dots are cell lysates. The right column are dots of the cellular supernatant. Note the faint dot adjacent to the dark lysate dot. This is a sign of successful secretion.
