The part codes for a synthetic luciferase and a NADPH-flavin oxidoreductase, joined by a 2A peptide sequence. When provided with exogenous substrate or coexpressed with the lux system fatty acid reductase complex, the part allows for light production. Luciferase catalyzes the conversion of substrate for light production, while the oxidoreductase regenerates FMNH2 for use by luciferase. The 2A peptide allows for stoichiometric expression of the two proteins. The part is comprised of two unique genes - monomeric LuxAB and frp. Each gene has an attached antibody tag - LuxAB has a MYC tag and frp has a HIS tag. Genes used are codon optimized for expression in yeast.
