Xylene Sensor! With AmilCP!
Constantly produces XylR and fluoresces in the presence of xylene. The Pu Promoter is activated when xylene becomes attached to XylR, which then produces AmilCP. Our experimental trials lead to positive results of this BioBrick part. Using the results we have found, this BioBrick is capable of measuring at least 100 PPM of xylene in a soil sample. When using experimental trials of xylene in LB Broth containing cells, we were able to detect 50 PPM. Further documentation is needed of this part to determine the extent to what amount of xylene this BioBrick can detect.
NOTE: Be prepared if using JM109 cells, as if put under stress, they can turn blue and lead to false results.
We recommend that the incubation of the bacteria last approximately 24 hours for optimal results.
