Red light sensing system with mRFP reporter
This part consists of a red light sensing system(BBa_K1797001) and an RFP reporter system. When bacterial colonies are exposed to red light, the OmpR promoter is not activated, preventing the expression of RFP. When colonies are kept in dark, the corresponding transcription factor of mpR promoter is activated and RFP is expressed, making the colonies red.
Part Demonstration
We transformed this part into E.coli and cultured bacteria in specialized tubes. One tube is treated with white light, since the wavelength of red light is included in white light. The other tube is wrapped in tin paper to avoid the illusion of blue light. After being cultured for about 17 hours, we compare the red color between the two tubes. From the following photo, we can see the significant difference in red color strength between two tubes. The observation that the tube kept in dark shows stronger red color is in accordance with our expectation, showing that our part actually works.
