RtTA-flag is a regulatory protein that can regulate the binding of tetracycline analogues.On the N side, a flag tag is brought.The purpose of the design is to verify that the rtTA regulatory proteins are able to be stably expressed in yeast cells, and the role of the project is to be verified.
rtTA is from the commercial plasmid pTet-on,flag tags is commonly used detective tags.Using the part as a test to verify whether the rtTA protein can be stably existed in the recombinant yeast cells, the flag tag is used to check the protein level using westernblot.We bought commercial plasmid pTet-on from biowit , and designed primers to obtain the target fragment from the plasmid through PCR technology, and then add the flag fragment by PCR technology.This is a chance for us to make further optimization and adjustment to make it more suitable for the host cells of Pichia pastoris, and the effect of flag fragment on rtTA protein is also unknown.
