Bistable switch 1 testing device
Bistable switch based on termosensitive lambda repressor (cIts) and lactose operon inhibitor (lacI).
Usage and Biology
The state of the switch is set either by IPTG or temperature. In 0,1 uM IPTG lacI gets inactivated ant transcription from Plac starts. It results in production of cIts and other genes belonging to this part of switch (in this case GFP). After removal of IPTG the state remains stable - PRm is repressed by cIts and genes under Plac are expressed. In 42oC cIts gets inactivated and genes under PRm (in this case lacI and RFP) are expressed. Because lacI is expressed to Plac gets repressed. The state remains stable after transfer of this device to 30oC. State of the device can be switched by pulses of IPTG and 42oC. Temperature of stable work is 30oC.
