M13 -47 general primer as a reverse primer binds to 5prime terminal of lacZ
Used as a internal primer in sequencing long composite BioBrick parts containing lacZ or lacZalpha.
Usage and Biology
SCUT-China 2023's Characterization
Paired with M13-48, they are a pair of primers on the plasmid pSET152PZ skeleton. As they are located on both sides of the NdeI cleavage site on the pSET152PZ plasmid, and the NotI site was used to construct the plasmid in this project, M13-47 and M13-48 are commonly used as universal primers for colony PCR in this project. The following gel plot shows the PCR results of the transformed colonies after inserting Prochi18B into the pSET152PZ plasmid. The M13-47 and M13-48 primers are expected to amplify a 508bp fragment, and according to the gel plot, the corresponding bacteria in lane 1 have been transferred into the successfully constructed vector.
