Circuit encoding leucine dehydrogenase and formate dehydrogenase
It is a complete device which could express two enzymes leucine dehydrogenase and formate dehydrogenase. When the reaction system was added NADH and trimethylpyruvic acid, leucine dehydrogenase converts the substrate into L-tert-leucine. At the same time, NADH is oxidized to NAD+. While the formate dehydrogenase can convert formate into carbon dioxide. NADH is regenerated. The conversion of TMP is 53.02% and the e.e value of product L-tert-leucine is >99%.
