Biosynthesis of beta-carotene
The plasmid contains the native promotor from the Pantoea ananatis gene cluster with the genes crtB, crtI, crtY and crtE in one operon and a kdgM terminator for the biosynthesis of beta-carotene in an operon design. This means that you need just one promoter to express all four genes that are needed for the biosynthesis. Furthermore, an origin of replication for E.coli is included. This origin of replication makes the plasmid high-copy. We decided for the BioBrick backbone with an Ampicillin resistance because we want to use it in the future in combination with another plasmid that carries Chloramphenicol resistance.
