J23100+Riborugulator+GFP

This part is constructed to test the function of promoter J23100 and riboregulator through the expression of GFP protein. This part is composed of a promoter, a riboregulator, an RBS, a protein coding sequence and a terminator. The promoter J23100 is a strong promoter, and it comes from the constitutive promoter family, these promoter parts can be used to tune the expression level of constitutively expressed parts. The NheI and AvrII restriction sites present within these promoter parts make them a scaffold for further modification. The riboregulator is lock 3 and can be opened by key 3 in our other part, the riboregulator can form a hairpin structure spontaneously when it is transcripted in the cell, and this structure can prevent ribosome binding to the mRNA, when its key - taRNA comes, this structure can be loosed. By this way, it can act as the regulator of mRNA expressing. The followed RBS+GFP+Terminator is an unit to test the function of the promoter and the riboregulator.
