bslA, forms hydrophobic surface layer on biofilm
To make our biofilm more robust and hydrophobic we we have focussed on bslA. bslA forms a hydrophobic surface layer on the biofilm of B. subtilis NCIB 3610 comI (ΔcomI). This layer is essential for the water repelling properties of the biofilm of the Bacillus subtilis. The BslA protein itself is a cell wall protein with amphiphilic properties which self-assembles into polymers at the air-water interface in vitro. For the expression of bslA, expression of epsABCDEFGHIJKLMNO and tapA-sipW-tasA operons are required. Without bslA the microstructure of the biofilm is smoothened and the repellency against water and organic solvents is reduced.
In our construct we added the salt inducible PproHpromoter (BBa_K1597000) before bslA before integration in B. subtilis. This strains was grown on Msgg medium with and without 0,5M NaCl for 24 hours. Images were taken with identical settings. The left image is without NaCl, where the right picture is with NaCl.
The blsA overproduction strain grown on Msgg without NaCl (left image) showed no clear phenotype compared with wildtype phenotype. However, when grown on 0,5M NaCl, ice crystal like structure were visible. This is futher described in tasA and bslA single and double overproduction strains.
