recA promoter (J22106) + LacZ gene (I732017): DNA damage sensor
This part combines the promoter of the recA gene (an SOS promoter) and the full LacZ gene, with Amp resistance from backbone plasmid pSB1A2. Original part numbers are J22106 (recA) and I732017 (lacZ). The promoter is in front of the LacZ gene, and therefore signals transcription of LacZ when the SOS DNA repair mechanism is triggered by massive DNA damage. LacZ encodes for beta-galactosidase, which cleaves lactose and its homolog X-gal. X-gal is cleaved into galactose and an insoluble blue dye. By plating on an X-gal plate, a bacterium with this plasmid should turn blue when exposed to extensive amounts of DNA damage.
