K1463100 RBS + GvpA + B0032 RBS + GvpC

A mutant version of the BBa_B0034 RBS (with the part name BBa_K1463100) upstream of GvpA BBa_K737016 followed by the BBa_B0032 RBS upstream of GvpC, BBa_K737017


Usage and Biology

This part was designed to express the two main structural proteins of gas vesicles from Planktothrix rubescens. The idea was to make E. coli float on expression of these genes. This isolate had a mutant version of the BBa_B0032 RBS in front of gvpA and probably does not express gvpA well. We suspect that over expression of gvpA and gvpC is toxic for the cell as we were unable to obtain the wild-type BBa_B0032 RBS in front of gvpA in pSB1C3. We were able to isolate the desired construct with BBa_B0032 driving gvpA expression, and BBa_B0034 driving gvpC expression downstream of a tightly regulated pBAD promoter.
