Blue Light Response Element

This part is a blue light response element that allows repression of the downstream gene expression with the intensity of blue light (around 470 nm). It consists of a medium strength Anderson promoter (BBa_J23106), YF1 and FixJ light receptor system coupled with strong RBSs (BBa_K592016), a double terminator (BBa_B0015), the FixK2 promoter (BBa_K592006) and a strong RBS (BBa_B0034). You can add the gene of your interest coupled with a terminator for the device to work.

Usage and Biology

According to Aalto-Helsinki team's (measurements), the blue light response element is capable of fourfould repression in less than 20 minutes (figure 1).

Figure 1: Blue light induced flourescence We incubated 9 bacterial cultures of 100ul in identical conditions in darkness. After approximately 1.5 hours of incubation, the samples were put under different intensities of light. The percieved intensity values are 10 base logarithms of the actual light intensities
