T7 promoter-driven BMC structural genes with a His6-tagged component for purification
Signal peptide for targeted Eut Bacterial Microcompartment localization
Usage and Biology
The signal peptide can be readily fused to any Biobrick proteins that have the start codon immediately downstream to the standard prefix, and can be ligated in via restriction with XbaI and PstI.
For example, after digesting this Biobrick in pSB1C3 with SpeI and PstI, and digest BBa_J06702 with XbaI and PstI, and ligate the two Biobricks together, one will get a fusion like this:
- Signal peptide-Thr-Gly-Ser-Val-Leu-Glu-mCherry
which would allow localization of the mCherry protein into the Eut-type of Bacterial Microcompartment. The Eut-type of Bacterial Microcompartment has also been designed and submitted by our team for in vivo and in vitro applcations as it is purifiable. For details please see BBa_K1428001 page on the registry.
