A combination of theophylline aptamer and taRNA that can response theophylline to regulate circuit

this part was made from the riboregulator taRNA by Isaacs et al(1)and theophylline aptamer by Lynch et al(2).Based on our desigen, the sequence of taRNA change some bases, while theophylline aptamer in our project is the same as Justin P.Gallivan group(2).

Usage and Biology

Fabrication notes:

In order to response difference molecular to regulate the gene circuit, we add theophylline aptamer based on the sequence of taRNA. When this piece is transcribed into mRNA,the aptamer's second structure complement with taRNA in term of the optimal configuration.When theophylline add into medium, aptamer can specificity combine it,changing the second structure,leading to the taRNA exposure.Now the key play a role to "wake up" our project lock. Part is also available with a constitutively on promoter as BBa_R0040, which is promoter tetR.
