LamB and TetA expression plasmid
Constitutively expresses the lamB gene, which codes for the lambda receptor. Following the lamB gene is the tetracycline resistance cassette.
Usage and Biology
The series of parts K137120-K137123 were placed in plasmid pSB1A2 and E. coli strain JW3996-1 (lamB::kanR). When infected with phage lambda, the cells produced the following relative plating efficiency (pfu/mL on engineered strain relative to pfu/mL on wild-type DH10B):
The plating efficiency correlates to expression level (determined using the promoter and RBS strengths listed in the Registry) with almost perfect linearity, suggesting that LamB expression is limiting infection frequency.
