rigid linker for λ-Lysozyme
This linker with the amino acid sequence GGAEAAAKEAAAKAAP was used to circularize λ-Lysozyme. The purpose of this linker is to achieve maximal thermostability, while preserving the native 3D structure of the protein. Our model http://2014.igem.org/Team:Heidelberg/Modeling/Linker_Modeling is based on the assumption that linkers have to be as short and rigid as possible in order to allow minimal conformational changes upon heating. On the other hand, linkers for circularization should be long enough not to cause conformational strain. This could impact both protein function and the cyclization reaction.
