CMV-SS-HA-multiHP-(TMTIR)TLR4-Histop

A synthetic Helicobacter pylori multiepitope antigen fused with the transmembrane (TM) and cytosolic (TIR) domains of human TLR4 under control of constitutive promotor CMV and equiped with HA-tag on N-terminus and His-tag on C-terminus.

Construct codes for a transmembrane protein expressed on the cell surface. It is possible to lable the protein with fluroscently labeled antibodies against the HA-tag on the outside of the cell or against the His-tag in the cytosol.

This part has shown constitutive homodimerization and activation, stimulating the innate immune response. The activation of this construct increases transcription of genes under NFkB promoter, which was measured by luciferase assays. After activation, the construct is internalised, the multiepitope processed inside endosomes and short peptide fragments presented on MHC II molecules. The antigen presented on the cell surface can be recognized by B-cells, which further increases immune response.
