Constitutive T7 promoter + Lock (crRNA)
This is the cis-repressive sequence that is downstream of a T7 promoter, which is a constitutive promoter that produces high levels of transcription in E. coli. A gene can be placed downstream of this part so that cis-repressed mRNA (crRNA) of the gene is constantly produced. However, the gene will not be expressed into proteins because the cis-sequence of the mRNA is complementary to the RBS. As a result, a stem loop formation occurs and the RBS is blocked off, preventing protein expression from occurring.
