pGlnA + hmsHFRS operon
Usage and Biology
To control the expression of the operon, we used a promoter from the standard registry of parts, which was characterized by our team last year (see Part:BBa_K763003).
The hmsHFRS operon encodes 4 proteins: protein HmsH goes from 211 to 2560 bp, protein HmsF from 2561 to 597 bp, protein HmsR from 4598 to 5968 bp and protein HmsS from 5969 to 6341 bp.
The molecular mechanism works as follows: with low nitrogen in the media, the promoter is activated and a biofilm of E.coli over C.elegans should be triggered,with high nitrogen concentration, the biobrick is inactive and the bacteria will not form the biofilm.

These plates are half-made of NGM media, which is poor in nitrogen, so it induces the formation of the biofilm by activating the GlnA promoter; and half-made of PHA media, which has fatty acids that induce the Clumping and the Building of the bioplastic (PHA).
