pTet_Lac hybrid promoter

This promoter is expected to be repressed by tetR and lacI. It contains two binding sites for TetR and one LacI binding site. The part is a predicted AND logic gate; transcription requires the presence of both aTc AND IPTG.

Summary: Because we want to use an pTet_ Lac hybrid promoter as a part of choice device in our project. So we do some research about several kinds of pTet_ Lac hybrid promoters’ characterization such as this(TTL) . The LTT promoter will have the more active GFP expression than this TTL promoter.

Conditions are ±100 ng/mL aTc and ±1mMIPTG, e.g., ± refers to 100 ng/mL aTc and 1mMIPTG. Here, the profile of the TTL system (a) is examined in detail at four points asmarked by the colored rings. Each of the four points circled by a colored ring in (a) corresponds to the identically colored histogram plot as depicted in panel (b). The histograms produced by the models, however, are free from the noise (dust, dead cell debris, etc.)
