This is the BioBrick-compatible backbone for pBSEP1, containing two origins of replication, one for gram-positive, and another for gram-negative bacteria. It has two selectable resistance markers, one for Chloramphenicol (5µg/ml) in Bacillus subtiis, and one for Ampicillin (100 µg/ml) in E. coli.
This vector can be used to construct large BioBrick parts using traditional gram-negative-based transformation and minipreps, and then also be used to hold the completed part for expression in a gram-positive chassis.
Note that unlike the image, this vector is empty and does not contain any other BioBricks between the cut sites.
As the figure explains, this vector was made via InFusion assembly from several pieces at once.
