This parts have been extracted from the LexA Operator Library.
The LexA repressor from Escherichia coli negatively regulates the transcription of about 20 different genes upon binding with variable affinity. Nature itself provide us a wide spectrum of operator sites with different activation thresholds.
K079039: Lex A1 is a perfectly 2-fold symmetrical consensus sequence binding LexA more tightly than any natural occurring SOS promoter. Published data show the details of this interaction but in vivo measurment of repression levels are still scarce
K079040: Lex A2 is the wild type operator sequence controlling RecA protein expression. Being the first activated response gene in the LexA degradation driven [http://2008.igem.org/Team:Bologna/Modeling#UV_Radiation:_SOS_system SOS Pathway] the respective operator have to be one of the least LexA affine natural sequence.
The above-mentioned operator selection should gurantee well distinct regulation dynamics and a sensitive response to different levels of Dna Damage, associated to qualitatively different phases of the Sos Pathway.
The parts can be individually assembled with Constitutive Promoter Family or combined since the binding of LexA to multiple operators cooperativity has been observed if the spacing between these operators is favorable.
