Protein Linker SSGSSG Library
Variant library of 6-amino-acid fusion protein linker sequences, encoded with different codon permutations while maintaining the same SSGSSG protein sequence, and the same codon usage count.
Sequences from this library can be picked from when designing a construct containing multiple fusion proteins (e.g. fusing GFP to protein A, RFP to protein B, YFP to protein C, BFP to for protein D, all in the same plasmid). Select a different linker for each fusion rather than reuse multiple copies of the same linker to reduce the chance of homologous recombination/misprimining event/gibson assembly overlap similarity.
Permutations of the codons are generated while following the rules:<br\> 1. encoding only SSGSSG<br\> 2. using only codons below, where codon frequency in E. coli is represented between [], and its symbolic representation between ()<br\> Serine: AGT(S)[8.7%], AGC(s)[16.1%], TCC($)[8.6%]<br\> Glycine: GGC(G)[29.8%], GGT(g)[24.9%]<br\> 3. all above codons can be only used once except for (s), which is used twice<br\> 4. no two of the same codon can be next to each other, in this case no (s)(s) should occur<br\>
