CH1 domain of human antibody + cleavage site + phoratoxin
Device is composed of four subunits. Primarily, the CH1 domain of human antibody(Part:BBa_J120020), a specific cleavage site (Part:BBa_J120023) that is recognized by the proteolytic function of ADAM17,phoratoxin(Part:BBa_J120022)and terminator (Part:BBa_B0011) . These four subunits are arranged in corresponding order. A promoter with a variable region (Part:BBa_J120021) must be linked to this device in order to function properly. For the antibody recognition to function properly, this part also needs the ADAM17 antibody variable region of light chain(Part:BBa_J120031) and kappa 1 immunoglobulin light chain constant region of human antibody (Part:BBa_J120021). These variable regions whose sequences are not known will recognize the ADAM17 present on the cell membrane and bind to it. The proteolytic function of ADAM17 will recognize the cleavage site and cleave it. This cleavage will result in releasing of phoratoxin which will lyse the cell membrane and eventually kill it. This device should be tested in a yeast cell, because glycosylation problems might occur in a prokaryote.
