RFP(rev)-RBS(rev)-pT7(rev)-pLac-RBS-T7RNAP
Expression of the T7 RNAP gene by the pLacI promoter (BBa_K091110) results in T7 RNA Polymerase, which drives production of RFP from the pT7 promoter. The RFP gene is backwards to prevent any readthrough transcription from the vector and to prevent runaway positive feedback from T7 RNAP expression. The T7 RNAP genes is engineered with two mechanisms for introduction of frameshift mutations for testing frameshift suppression. One uses the two 8 bp recognition enzymes AsiSI and AscI and the other uses BsaI automated assembly.
