LuxR-repressible promoter library member
This promoter contains a lux box in the core region that implements a repressible regulatory behaviour in presence of LuxR and AHL. It is part of a library, summarized below, including promoters with different -35 and -10 sequences, thus resulting in a different transcriptional activity. The characterization data provided here are relative to promoters with the BBa_I13507 RFP expression system downstream (with the TACTAGTG scar between promoter and RBS) in the pSB4C5 plasmid, tested in TOP10 in the M9 medium supplemented with casamino acids and glycerol in a microplate reader. Activity values are expressed in RPUs. Characterization was also carried out in other contexts, reported in the publication, such as different plasmids, reporter genes, measurement equipment and LuxR sources.

Characterization with LuxR (via the co-transformed BBa_S03119 in the pSB3K3 plasmid) as a function of AHL:
Experimental data were fitted with the Hill function RPU=delta + Vmax/(1+(AHL/Km)^eta) and estimated parameters are reported for each library member.
