J100287 with two bases removed between gene III and GFP
J100287 is a plasmid that could be used in the positive selection phase of Phage Assisted Continuous Evolution (PACE) to evolve riboswitches. Gene III, the gene necessary for phage survival, and GFP, a reporter gene, are separated by a stop codon at the end of gene III. J100310 had two bases, specifically "TA", in the stop codon between the genes removed, forming a translational fusion of the two proteins. While both gene III and GFP should be produced in J100287 since the GFP gene has its own ribosome binding site, J100310 is another option to try for PACE.
