nifB promoter

The nifB promoter comes from the Paenibacillus that lives in bamboo. The promoter is always turned on until repressed by the presence of nitrogen. The gene is involved in nitrogen fixation. We found a paper that includes the nifB promoter on pubmed, and planned to make an oligo with no more than 60 base pairs and created the correct sticky ends to work with Golden Gate Assembly. The authors of the article hypothesized that the promoter would be repressed by ammonium chloride but never confirmed with testing, so we decided to test this hypotheses. The oligo will be placed in the presence of RFP to see if it will glow red.
