Ste2-GFP-WMTM1 under endogenous promoter

Usage and Biology

Ste2 is a native to the yeast Saccharomyces cerevisiae, and is an integral membrane receptor for the mating pheromone alpha factor. Upon induction with alpha factor, Ste2 is endocytosed into the cell, and eventually ends up in the vacuole.

WMTM1 stands for Worm (C. Elegans) Myotubularin(MTM) related protein 1. WMTM1 binds Phosphatidylinositol 3-Phosphate (PI[3]P) and Phospatidylinosotol 3,5-Phosphate (PI[3,5]P) as substrates and dephosphorylates them at the 3-Phosphate position.

This part has been tested, and we believe that Ste2 is being mislocalized and is not functional in the native pathway. In addition, its phenotype exhibits a much larger vacuole as compared to wild type yeast.
