cI (+LVA) repressor regulated by pLac Promoter, GFP (+LVA)

IPTG presence detector and cI inducible expression. In presence of lactose or IPTG, an analog of lactose, LacI is unable to correctly bind and inhibits transcription. This allows BBa_R0010 to be used as a detector of lactose or IPTG thanks to the development of green florescence for the presence of GFP coding sequence (J04031). the GFP is modified with an LVA tail for rapid degradation of the protein. The presence of IPTG induces the expression of cI repressor (C0051) modified with an LVA tail for rapid degradation of the protein. cI repressor binds to the cI regulator (BBa_R0051). It can be used as a detector of lactose metabolism.
