Outer Membrane Permeability Device
This device employs the FepA L8T mutant described here to allow molecules larger than normal to pass through the outer membrane of E. coli. The porins usually present will allow molecules up to around 700Da to freely diffuse in and out, however with this device, molecules of at least 1400Da should be able to pass through. Permeability was tested using the antibiotic Bacitracin. Wild type E. coli are resistant to this as the molecule is 1422Da in size. Expressing the device made the bacteria sensitive to bacitracin. In this device, the mutant FepA is under the control of a relatively high strength promotor.
