The artificial repressor comes from the results by [http://2007.igem.org/USTC/Repressor_Evolution_on_Plates Repressor Evolution on Plates]. The natural lactose repressor protein has been redesigned at Position 17, 18 of a.a. sequence, which is the composition of amino acid of recognition helix (Figure I). Standard BioBrick Prefix and Suffix are added to both ends of this part for standard assembly.
Usage and Biology
- This artificial lac-repressor can bind to the specific promoters with high affinity while weakly bind to other promoters with relatively low affinity. The repression intensity is indicated in the Repression Matrix shown as below. - This repressor sequence is single repressor expression parts without other functional gene parts such as RBS & Terminators. The same repressor sequence with RBS can be found in BBa_I732807 as a Standard BioBrick part. The same repressor sequence with both RBS and Terminator can be found in BBa_I732827 as a Standard BioBrick part.
