CMV promoter - Myc tag - CCR5 (shortened) - TEV N
Human CCR5 shortened receptor fused with N-terminal half of TEV protease and Myc tag. This construct is under control of CMV promoter.
Human CCR5 transmembrane receptor is a chemokine receptor which plays cruical role in HIV infection. This version of CCR5 is shortened for 50 aminoacids from C-terminus, therefore is without cytosolic domain. Myc tag is fused to the N-terminal of this protein.
CCR5 (shortened) is also fused with N-terminal half of TEV protease. This construct is coexpressed with CD4 receptor fused with C-terminal half of TEV protease (in mammalian cells). When that kind of human cells are infected with HIV virus both halves of TEV protease assemble and split protein regains its function. This principle can be used for fighting HIV infection.
