E. colibrator design 3 has the OR gate embedded within the switch via a double instance of I3400 Hk022cI switch input device. Design 3 has an aiiA mechanism turn on during the switch OFF state, as illustrated by I0460 AiiA device immediately following I3411 Lambda cI switch output device. This allows the OFF state to speed up reset of the system by degrading residual HSL once quorum threshold is reached.

Usage and Biology

The E. colibrator system is designed to create dynamic polkadots in E. coli. Cells are in one of two states stored in a bistable switch. One state, triggered by a stochastic Lac trigger or by the chemotaxis aspartate molecule, turns on a CFP reporter gene and causes synthesis and excretion of aspartate. The other state, triggered by quorum sensing under dense conditions, turns off the reporter gene and aspartate secretion and turns on aiiA synthesis.
