Adaptable Vaccine Antigen Secretor

Bacteria secrete a variety of biological agents in order to function and perform tasks. This device is designed so that the E. Coli will be able to export any protein structure whose genomic sequence is inserted into the device. The device is specifically designed so that secretion can be prevented in vitro. A series of promoters is used to ensure that the device is turned off in vitro if the media contains atc. Atc activates the Tetr promoter, which causes the translation of lac5. Lac5 blocks the activation of the lacI promoter which must be activated to begin transcription of the ribosome binding site, secretor sequence etc. This is specifically designed so that the secretor function will be activated in vitro. The actual secretor sequence has been demonstrated to work in E. Coli. The secretor sequence is connected to the desired secreted agent via a glc/ser linker. This linker has been codon optimized for E. coli.
