3OC6HSL amplifier

This device is a strong amplifier of 3OC6HSL. R0065 is a leaky promoter that produces enough LuxI to generate ample HSL to switch this device to a stable high expression state at a particular cell density.

Usage and Biology

Expression from R0065 is leaky, causing this device to demonstrate typical quorum sensing behavior (density dependent activation). The circuit switches from a discreet OFF to a discreet ON state at a cell density which is dependent on the pH of the media (~0.9x10^9 cfu/mL in LB pH 7.0). The circuit can be kept in the OFF state by TetR, and induced to different extents by the addition of anhydrotetracycline. The circuit can also be dominantly repressed by expression of cI (lambda repressor protein).
