Design Notes

Primers used in the PCR: 

X174_E_FP: gcttctag atggtacgctggactttgt (TM:55C)
X174_E_RP: gct ctgcagcggccgctactagta tcactccttccgcacg (TM:55C)

Digested the PCR product with XbaI and PstI, and ligated the digestion product with a gel purified XbaI,PstI digested pSB1A2 plasmid backbone.
