Applications of BBa_K546003

This part serves as a weak constitutive producer of luxR. luxR can form a complex with 3OC6HSL (AHL) inducing the pR promoter.

This part was tested in conjunction with BBa_K546002 and is the repaired F2621. The F2621 was sequenced by our team and appeared to miss the R0063 promoter. K546003 thus corresponds to F2621 repaired by R0063 and was tested with GFP as a reporter. This was also done for the 'broken' version. The parts' responses to AHL were tested by adding 0, 250 and 1000 nM AHL.

The fluorescence measurements are depicted in the diagram underneath. The detection limit of the apparatus was 13000 RFU for this experiment, as is visible in the diagram. R0063-F2621-GFP refers to this part and clearly responds to AHL as the GFP expression rapidly increases over time. F2621-GFP refers to the broken version of F2621 (thus without the constituve luxR because R0063 was missing in this part). This broken version hardly responds to AHL.
