PhaJ (Hydratase)
PhaJ ((R)-Specific enoyl-CoA hydratases) catalyzes the oxidation of enoyl-CoA to (R)-3-hydroxyacyl-CoA. Where (R)-3-Hydroxyacyl-CoA is the substrate for PhaC and the direct precursor of PHA biosynthesis. This occurs if the carbon source is oxidized through the fatty acid b-oxidation pathway (Tsuge et al., 2015). This enzyme converts trans-2-enoyl-CoA into (R)-3-hydroxyacyl-CoA and uses an intermediate originating from the beta oxidation cycle used for the metabolism of fatty acids. (R)-3-hydroxyacyl-CoA monomers can be polymerized into polyhydroxybutyrate (PHB) using PhaC (Sato et al., 2011). This enzyme is involved in the biosynthesis of PHB with an approximate mass of 18kDa.
