Pro/Thr-rich Linker

The Pro/Thr-rich linker is one type of rigid linker. Different from the flexible GS-Linker, the Pro-rich linker doesn't have the alpha helix. The rich proline amino acid in the linker can increase the hardness of the backbone, thus can separate protein domains efficiently. By changing the number of Thr-Pro sequences, we can easily change the distance between the two domains, so that we can ensure the activity and biological function of proteins on both sides of the Pro/Thr-rich linker.

In addition, the Pro-Thr box was found on the conjugated sequence between the catalytic domain and cellulose-binding domain(CBD) of endoglucanase. Deletion of the Pro/Thr box alters the conformation of the endoglucanase by changing the relative orientation of the catalytic domain and the CBD. The deletion does not affect the adsorption of the enzyme to microcrystalline cellulose, but it does affect its desorption from cellulose.
