Usage and Biology

This part will express the valencene synthase, which catalyzes the conversion of FPP to linear and cyclic sesquiterpenes. The chemical equation is(2E,6E)-farnesyl diphosphate = (+)-valencene + diphosphate. The reaction product (+)-valencene can be oxidized to produce our target product (+)-nootkatone.

In our project, VS will work with HPO (BBa_K3772035), AtCPR (BBa_K3772036), ADH3 (BBa_K3772037) and other endogenous enzyme systems of S. cerevisiae CEN.PK2-1Ca to synthesize nootkatone.


Fermentation production of nootkatone

The modified engineered strain S. cerevisiae CEN.PK2-1Ca was inoculated in 10 mL of SD medium system deficient in tryptophan, uracil and leucine. And the product nootkatone was assayed after incubation at 25 ℃, 200 rpm for 96h.
