Description

This part contains the ribosomal binding site B0034 and riboJ which is a ribozyme. RiboJ comprises the ribozyme with an additional hairpin to help expose the RBS. RiboJ contains both the ribozyme and hairpin (Lou, Stanton, Chen, Munsky, & Voigt, 2012).

Upon translation of the ribozyme into RNA, it cleaves itself off, removing any transcript from the mRNA upstream of the ribosomal binding site (RBS).

Figure 1: Maturation of mRNA transcripts leads to a more standardized genetic context.

This leads to a more stable expression independent of the cellular context (See iGEM William and Mary 2016) and can be used for better cross comparison of results over species border or the construction of more robust, species independent functional devices.

Compability

These parts are compatible with the RCF [1000] standard and can be used in any part collection that uses the PhytoBrick standard of overhangs. For more information we recommend to head over to Design of the Marburg Collection.
