Vibrio natriegens' TorA signal peptide containing human growth hormone with 6xHis
This is a composite part containing Vibrio natriegens' TorA Tat signal peptide (BBa_K3016100) fused with human growth hormone encoding gene (hGH, BBa_K3016500), including 6xHis tag in the C-terminus of hGH.
Usage
The fusion with TorA signal peptide sequence (ssTorA) enables the translocation of hGH to the periplasmic space in Vibrio natriegens via the twin-arginine translocation pathway (Tat pathway). Purification of the protein from periplasmic space is advantageous e.g. due to lower amount of contaminants. His tag in the C-terminus enables easy purification or detection of the protein product.
Biology of the human growth hormone (hGH)
hGH is a peptide hormone that stimulates growth, cell reproduction, and cell regeneration in humans, and is essential in human development. It also stimulates the production of IGF-1 and increases the concentration of glucose and free fatty acids in plasma.
hGH is a 191-amino acid, single-chain polypeptide. In humans it is synthesized, stored and secreted by somatotropic cells located in the lateral wings of the anterior pituitary gland. hGH has a molecular weight of 22,124 Da. The protein structure includes four helices that interact with the GH receptor. The genes that encode human growth hormone are growth hormone 1 (somatotropin; pituitary growth hormone) and growth hormone 2 (placental growth hormone; growth hormone variant), and they are located in the q22-24 region of chromosome 17.
Clinical significance
A recombinant form of hGH is used to treat growth disorders and growth deficiencies. These include Turner syndrome, Prader–Willi syndrome, and chronic renal failure.
