Applications of BBa_K137018

This part is meant to measure the response of the part F2622 to induction with acylhomoserine lactone (AHL). By measuring GFP flourescence on a flow cytometer, the ability of F2622 to activate transcription of a target gene can be measured.

Lux R is expressed upon induction by isopropyl-b-D-thiogalactopyranoside (IPTG).The acyl-homoserine lactone (AHL) signaling molecule can bind and active the Lux R transcriptional regulator, which induces the expression of GFP under the control of lux pR.

The figure below shows the performance of E.coli strain BL21 with the biobrick K137018 when exposed to ultraviolet light. IPTG and AHL are added to the medium. The concentrations of the signaling molecules(AHL) are 0, 2, 4, 6, 8, 10nmol, respectively.

From the figure, we can see that the higher the concentration of the signaling molecules(AHL), the stronger the fluorescence intensity.
