Transcriptional terminator

  • Terminator to be placed downstream of multiple cloning site in new pSB series plasmids. Similar to BBa_B0051.
  • Taken from Lucigen's pSMART VC vector (http://www.lucigen.com/products/Cloning_Systems/CopyRight.html). See NCBS accession number AY643800. Stem loop was identified using mFOLD. According to Lucigen, this flanking terminator enabled cloning of strong promoters which failed to clone in other vectors. Note that this sequence may be somewhat longer than what is strictly necessary for transcript termination. I was conservative in my design of this part (i.e. I was generous in how much sequence I took) in the hopes of not accidentally adversely affecting termination efficiency.

This terminator is derived from bases 1309832 to 1309871 of E. coli MG1655 accession U00096. This is the intergenic region between genes tonB and yciA. This terminator is likely bidirectional since it is in between tonB (forward direction) and yciA (reverse direction).
