YlARO3 (ARO3 gene derived from Yarrowia lipolytica)

Biology and Usage

The YIARO3 gene is used to relieve the inhibition of L-Phe and L-Tyr on the activity of the DAHPS enzyme. DAHPS enzyme plays a vital role in the shikimate pathway and is responsible for the synthesis of chorismate. Once the activity of the DAHPS enzyme is inhibited by L-Phe and L-Tyr, the carbon flux into the shikimic acid pathway will be significantly reduced. By introducing the ARO3 gene, we eliminated the feedback inhibition of L-Phe and L-Tyr on DAHPS enzyme activity, thereby increasing the carbon flux of the shikimic acid pathway to expect higher resveratrol production. To improve the efficiency of DAHP, we optimized the codon to adapt it to the N.benthamiana system. Based on the original sequence, the 157th aspartic acid was changed to asparagine to obtain ARO3 [ D157N ].
