GjALDH2C3_opt (aldehyde dehydrogenase)
Aldehyde dehydrogenase (ALDH2C3) was used in this project, which will catalyze the conversion of crocin dialdehyde to crocin [1]. Gardenia is used in traditional Chinese medicine and is of interest as a rich source of crocin [3]. The ALDH family is widely distributed in plants and can oxidize aldehyde oxides to the corresponding carboxylic acid compounds. However, ALDH shows strong specificity in catalyzing crocin dialdehyde [2]; as an essential step in the synthesis pathway, the auxiliary role of ALDH is vital, and it can significantly enhance crocin accumulation. Therefore, we hope to introduce this gene to improve the accumulation of crocin while perfecting the crocin synthesis pathway. ALDH2C3 was detected to be highly expressed in the red fruits of Gardenia jasminoides. In a study, the authors introduced the aldehyde dehydrogenase gene GjALDH2C3 and other genes related to carotenoid synthesis. They produced crocin at a maximum level of 78,362 ng/g FW in Hon's tobacco [2].
